A Non-Destructive, Tuneable Method to Isolate Live Cells for High-Speed AFM Analysis

چکیده

Suitable immobilisation of microorganisms and single cells is key for high-resolution topographical imaging study mechanical properties with atomic force microscopy (AFM) under physiologically relevant conditions. Sample preparation techniques must be able to withstand the forces exerted by Z range-limited cantilever tip, not negatively affect sample surface data acquisition. Here, we describe an inherently flexible methodology, utilising three-dimensional based printing technique multiphoton polymerisation rapidly generate bespoke arrays cellular AFM analysis. As example, present collected from live Emiliania huxleyi cells, unicellular microalgae, imaged contact mode High-Speed Atomic Force Microscopy (HS-AFM), including one cell that was continuously over 90 min.

High-speed AFM imaging.

Proteins are dynamic in nature and function at the single molecule level. To achieve a straightforward and in-depth understanding of their underlying functional mechanism, we need to directly observe protein molecules at work at high resolution, without the use of protein-attached markers. To realize such objectives, high-speed atomic force microscopy (HS-AFM) has been developed and recently it...

Leaf area estimation by a simple and non-destructive method

In this study, the relation between leaf area and its dimensions was estimated using a non-destructive method. This method is based on this fact that the leaf shape does not change during the growing season. In this method, leaf area during the growing season is estimated based on the dimensions of the smallest leaf in the initial stage of plant growth or at any growth stage by measuring the le...

presenting a new method for high speed incremental forming

recently, incremental sheet metal forming (ismf) process as one of the new methods in rapid prototyping collection has been considered by researchers. this process is based on a defined path in cnc controller and applies a spherical-head tool that supplies the required pressure for sheet metal forming. despite affirmation of process ability in forming symmetric geometries, some of its aspects s...

High-speed AFM and applications to biomolecular systems.

Directly observing individual protein molecules in action at high spatiotemporal resolution has long been a holy grail for biological science. This is because we long have had to infer how proteins function from the static snapshots of their structures and dynamic behavior of optical makers attached to the molecules. This limitation has recently been removed to a large extent by the materializa...

A method of applying Fourier analysis to high-speed laryngoscopy.

A new method for analysis of digital high-speed recordings of vocal-fold vibrations is presented. The method is based on the extraction of light-intensity time sequences from consecutive images, which in turn are Fourier transformed. The spectra thus acquired can be displayed in four different modes, each having its own benefits. When applied to the larynx, the method visualizes oscillations in...
